Resveratrol and sirtuins: the scientific debate
Contested mechanism, based on lab assays and yeast; no human longevity evidence.

Editorial section. Research summaries are not product claims and contain no product links or offers. A summary is never proof that any product works.
- The question
- Does resveratrol directly activate SIRT1, the 'longevity enzyme'?
- Study design
- Biochemical and yeast studies from 2003 to 2013 by competing labs, including David Sinclair's group, Matt Kaeberlein and colleagues, and a Pfizer team. [1][2][3][4]
- What they found
- In 2003, Sinclair's lab reported that resveratrol activated sirtuins and extended yeast lifespan by 70%. In 2005, Kaeberlein and colleagues found the activation appeared only with a fluorescent tag (Fluor de Lys) attached to the test peptide and saw no lifespan effect in three yeast strains. In 2010, Pfizer scientists reported that resveratrol and related compounds did not activate SIRT1 with native substrates. In 2013, Sinclair's group reported that activation can occur through a specific allosteric site when substrates carry certain hydrophobic motifs. Pharma interest cooled: GSK bought Sirtris, the company built on this work, in 2008 and later wound it down. [1][2][3][4]
- Limitations
- The debate is about mechanism in test tubes and simple organisms. None of these papers tested longevity in people.
- What this does not mean
- A contested mechanism is not proof of benefit or of harm. We do not sell resveratrol.
